Acyl adenylates; the synthesis and properties of adenyl acetate.

چکیده
A study of the mechanism of acetyl CoAl formation from ATP, acetate, and CoA by yeast aceto-CoA-kinase has revealed that the reaction occurs in two steps (1 , 2). The evidence suggests that ATP and acetate react to form PP and the acetyl derivative of A5P, which is then utilized in the formation of acetyl CoA. This formulation is supported by the finding that synthetic adenyl acetate and PP are enzymatically converted to ATP (Reaction I), and that acetyl CoA is formed from adenyl acetate and CoA (Reaction 2).

Several reactions are now known which can account for the exchange of PI” with the terminal pyrophosphate group of ATP (1-6). Recently it was demonstrated (3, 4) that the reaction of ATP and acetate with the acetate-activating enzyme (aceto-CoA-kinase) resulted in such an exchange. Because of this and other observations (4), it was proposed that the acetatedependent exchange occurred via the in...

This mechanism of amino acid activation is analogous to that first proposed by Berg for the carboxyl activation of acetate and methionine by yeast extracts.4 It is consistent with the requirement of an amino acid for pyrophosphate exchange into ATP,2-7 the failure of AMP to exchange into ATP,1 I and the transfer of 018 from the carboxyl group of the amino acid to AMP in the over-all reaction (s...

In previous communications from this laboratory (1,2), direct evidence for the enzymatic synthesis of tryptophanyl adenylate from adenosine 5’triphosphate and tryptophan by purified beef pancreas tryptophan-activating enzyme was reported. Similar findings have been published subsequently by Kingdon et al. (3). Earlier work (4-8) also provided support for the belief that activation of amino acid...
متن کاملSynthesis of Adenosine 5'- (Aminoalkyl phosphonates and phosphinates) Analogues of Aminoacyl Adenylates
Translation of the genetic code and protein synthesis from amino acids takes place in the ribosome’s where tRNAs act as key intermediates. Correct translation of genetic information into the specific amino acids is dependent on aminoacyl-tRNA ́s. The synthesis of these is catalyzed by aminoacyl-tRNA synthetase. The aminoacylation reaction, the so-called charging of tRNA is carried out in two ste...
